Protein Crystallization
The Crystal Tube Kit allows protein crystallization by capillary counterdiffusion. The counterdiffusion procedure consists of volumes of protein solution and precipitant solution arranged in juxtaposition against each other in a capillary x-ray tube. Unlike conventional methods, counterdiffusion allows screening of a range of crystallization conditions using a temporal-spatial gradient of supersaturation conditions along the length of the capillary tube. Postsoaking of the crystals in solutions containing ligands, heavy metals, or cryoprotectant is achieved by exchanging precipitant solution for postsoaking solution. The protein crystal environments can be changed without drastic chemical changes or physical manipulation. The Crystal Tube Kit is designed for use with a small amount of protein sample (2–10 µL) and is suited for high-throughput protein crystallization and quick crystal screening.
